    RelA/NF-kappaB recruitment on the bax gene promoter antagonizes p73-dependent apoptosis in costimulated T cells

    The balance between antiapoptotic and proapoptotic proteins of the Bcl-2 family is critical in determining the fate of T cells in response to death stimuli. Proapoptotic genes, such as bax, are generally regulated by the p53 family of transcription factors, whereas NF-kappaB subunits can activate the transcription of antiapoptotic Bcl-2 members. Here, we show that CD28 activation protects memory T cells from irradiation-induced apoptosis by both upregulating bcl-xL and inhibiting bax gene expression. We found that p73, but not p53, binds to and trans-activates the bax gene promoter in irradiated T cells. The activation of RelA/NF-kappaB subunit in CD28 costimulated T cells and its binding onto the bax gene promoter results in suppression of bax transcription and decrease in both p73 and RNA polymerase II recruitment in vivo. RelA recruitment on the bax gene promoter is also accompanied by the lost of p300 binding and the parallel appearance of histone deacetylase-1-containing complexes. These findings identify RelA/NF-kappaB as a critical regulator of T-cell survival by affecting the balance of Bcl-2 family members

    Non-linear coupled approach for the evaluation of seismic slope displacements

    The design procedures to evaluate earthquake-induced sliding displacements typically refer to three different approaches: simplified methods; displacements methods and advanced dynamic methods. In the first class of methods, empirical relationships are used to predict the permanent slope displacement. The second class includes simplified dynamic analysis, by means of the conventional Newmark rigid block model, as well as through its improvements to account for soil deformability. The dynamic site response and the sliding block displacements are computed separately in the “decoupled” approach or simultaneously in the “coupled” analysis (stick-slip model). The advanced dynamic methods are based on finite element (FEM) or finite difference (FDM) formulations, which permit to account for topography and heterogeneity by two or three dimensional analysis. The paper describes the developments of a 1D lumped-mass stick-slip model for a layered subsoil including more generalized assumptions than a previous version (Ausilio et al., 2008). The depth of the sliding surface is considered not necessarily coincident with that of the bedrock, and located in a generic layer. This latter can be identified during the analysis. In the non-linear site response analysis, a recent soil damping formulation was used (Phillips & Hashash, 2009). In this formulation, a reduction factor modifying the extended Masing loading/unloading strain-stress relationship was introduced. The predictions of the coupled stick-slip model with non-linear soil behaviour were compared with the results of the equivalent linear approach for ideal slopes to show the effects of non linearity on seismic performance. Besides, the results show that the sliding surface depth, automatically researched, is function of main ground motion parameters

    Beyond the fundamental noise limit in coherent optical fiber links

    It is well known that temperature variations and acoustic noise affect ultrastable frequency dissemination along optical fiber. Active stabilization techniques are in general adopted to compensate for the fiber-induced phase noise. However, despite this compensation, the ultimate link performances remain limited by the so called delay-unsuppressed fiber noise that is related to the propagation delay of the light in the fiber. In this paper, we demonstrate a data post-processing approach which enables us to overcome this limit. We implement a subtraction algorithm between the optical signal delivered at the remote link end and the round-trip signal. In this way, a 6 dB improvement beyond the fundamental limit imposed by delay-unsuppressed noise is obtained. This result enhances the resolution of possible comparisons between remote optical clocks by a factor of 2. We confirm the theoretical prediction with experimental data obtained on a 47 km metropolitan fiber link, and propose how to extend this method for frequency dissemination purposes as well

    Adaptive modulation control for visible light communication systems

    Visible light communication (VLC) builds on the dual use of lightening infrastructure for communication. Even though the advantages of VLC are well known, as emerging communication paradigm, some open issues still need to be addressed in order to rely on it as a robust communication system. First of all, external interference as an extremely varying signal impacting on the reliability of the VLC system needs to be analyzed. In this paper, we propose a system where the link conditions (in terms of signal-to-noise-ratio (SNR)) drive the modulation scheme and this procedure is managed through the use of an uplink/channel, to assure a feedback path. The receiver is in charge of choosing the modulation scheme matching the requirement in terms of error rate on the basis of the measured SNR after noise mitigation. The feasibility of the system and its effectiveness are evaluated by designing and implementing a complete bi-directional system. In particular, an uplink channel sending the information regarding the specific selected modulation technique has been implemented and the whole system is based on a fine synchronization approach in order to “track” in real time the most suitable modulation scheme. Experimental results show the effectiveness of a bi-directional system in order to implement an adaptive VLC system able to follow the environmental changes (in terms of interference and noise)

    Sustainability along the value chain: collaborative approaches and their impact on firm performance

    Heeding the call for a deeper understanding of the key differences in corporate approaches to sustainable supply chain management (SSCM) and their impact on performance, we collect and analyse data on a sample of 91 Italian firms in the consumer product industry engaged in upstream and downstream sustainability initiatives. Results show that companies differ in the extent to which sustainability is shared along the chain. Yet, the more companies collaborate through a strong sustainability orientation, the higher the impact on supply-chain related performance, with companies adopting a proactive attitude to SSCM being able to benefit the most from it

    Use of biological drugs in patients with psoriasis and psoriatic arthritis in italy: Results from the PSONG survey

    This Italian multicenter retrospective study compared the drug survival and efficacy of differentanti-TNF agents in psoriasis (PsO) and psoriatic arthritis (PsA) patients. A database of PsO/PsApatients treated with adalimumab, etanercept, and infliximab from May 2013 to May 2014 wasanalyzed. PASI 75, 90, and 100 was calculated at each time point to evaluate efficacy. Drug sur-vival rate and probability of maintaining PASI response were evaluated. The impact of dependentvariables on probability of PASI 75 loss was evaluated by logistic regression. 1,235 patients wereincluded, 577 with PsO and 658 with PsA. Highest survival rates were observed with adalimumabfollowed by etanercept and infliximab in PsO and PsA patients. The probability of maintainingPASI response was significantly higher for adalimumab followed by infliximab. For PsO patients,the odds of losing PASI 75 was higher in etanercept-treated patients (OR: 8.1; 95% CI: 4.2–15.6,p<.001) or infliximab (OR: 6.6; 95% CI: 2.6–16.3,p<.001) vs. adalimumab. Likewise, for PsApatients the odds of losing PASI 75 was higher in etanercept-treated patients (OR: 2.3; 95% CI:1.4–3.8,p5.01) or infliximab (OR: 2.2; 95% CI: 1.1–4.1,p5.018) vs. adalimumab. Adalimumabcould be the best therapeutic option over other anti-TNF agents for the treatment of PsO and PsApatients
